Schedule

Week 1 (April 1–5)

Lecture 1: Introduction to the course, lab safety, lab notebooks, the goals of lab, scientific manuscripts.

Lab

• Mon/Tues: No lab.
• Wed/Thurs: (0) Notebooks and lab safety orientation with TAs.

Assignments/Due Dates:

• Complete Lab Safety Fundamentals Training, and upload screenshot of worksafe transcript showing completion, before Lab 1.
• Complete the initial course survey on Bruin Learn.
• Sign up for a paper or equipment presentation.
• Asynchronous lecture on introduction to / purpose of lab.

Week 2 (April 8–12)

Lecture 2: Discussion of Ben-David et al.

Lab

• Mon/Tues: (1) Introduction to lab, measuring and microscopy.
• Wed/Thurs: (2) Replica molding and laser cutting demo, fluorescence microscopy.

Assignments/Due Dates:

• Read paper and answer reading questions before lecture.
• Asynchronous lecture on cell culture.

Week 3 (April 15–19)

Lecture 3: Discussion of Reticker-Flynn et al.

Lab

• Mon/Tues: (3) Bioconjugation.
• Wed/Thurs: (4) Microcontact printing, fluorescent microscopy of patterned proteins, and contact angle measurements.

Assignments/Due Dates:

• Read paper and answer reading questions before lecture.
• Asynchronous lecture on bioconjugation.

Week 4 (April 22–26)

Lecture 4: No lecture.

Lab

• Mon/Tues: (5) Cell culture—Passaging and counting cells, introduction to sterile technique.
• Wed/Thurs: (6) Cell culture—Growth kinetics, cell counting (hemocytometer), observing cell density and gauging confluence.

Assignments/Due Dates:

• Report for labs 1–4 due Thursday at 11:59 pm.

Week 5 (April 29–May 3)

Lecture 5: Discussion of Shaffer et al.

Lab

• Mon/Tues: (7) Growth kinetics, cell counting, lysis, protein content (BCA assay).
• Wed/Thurs: (8) Growth kinetics, MTS assay.

Assignments/Due Dates:

• Read paper and answer reading questions before lecture.
• Take home Exam 1.
• Asynchronous lecture on statistics.

Week 6 (May 6–10)

Lecture 6: Discussion of Zhang et al.

Lab

• Mon/Tues: Cancelled due to remote instruction.

Week 7 (May 13–17)

Lecture 7: Discussion of Engler et al.

Lab

• Mon/Tues: (9) 3D encapsulation, preparing substrates for cell studies.
• Wed/Thurs: (10) Live/dead, MTS assay on encapsulated cells, sterilizing substrates.

Assignments/Due Dates:

• Read paper and answer reading questions before lecture.
• Asynchronous lecture on mechanics.

Week 8 (May 20–24)

Lecture 8: Discussion of Fraley et al.

Lab

• Mon/Tues: (11) Varying stiffness substrates and cell growth, seeding cells.
• Wed/Thurs: (12) Quantification and analysis of cell growth and morphology.

Assignments/Due Dates:

• Read paper and answer reading questions before lecture.
• Asynchronous lecture on regression

Week 9 (May 27–31)

Lecture 9: Memorial Day. No lecture.

Lab

• Mon/Tues: Memorial Day, no lab. Backup lab session if needed.
• Wed/Thurs: (13) Lab Practical Exam.

Week 10 (June 3–7)

Lecture 10: Discussion of Sun et al.

Lab

• Mon/Tues: (16) Lab clean-up.
• Wed/Thurs: (17) Instron demo.

• Take home Exam 2.
• Report for labs 9-12 due on Thursday at 11:59 pm.
